The Aerosoft A320/21 is a thorough and complex aircraft. Unlike many others, if you are doing even the smallest thing wrong (procedurally), it may prevent other functions working, just like in the real aircraft. Don't worry, I am not accusing you of this, but it is something to keep in mind. The best place to start is the thorough PDF files that come with the product, i.e: tutorials, guides, etc.
The second best place (arguably first) is YouTube. Simply search "Aerosoft A320 Autopilot Tutorial" - plenty of great videos and guides.
It is hard to say what you are doing wrong without seeing it first hand, but just remember: on the FCU (Flight Control Unit - i.e: the autopilot control panel on the glareshield), by "pressing" the knobs you are "managing" them. What this means is that it will get it's orders from whatever you have programmed in the CDU (Control Display Unit). If you "pull" the knobs, you are "un-managing" them. This means that you can then adjust (twist) the knobs to set headings, altitudes, vertical speed, etc.
